Wachstumsfaktoren (FGF)

Die Familie der Fibroblasten-Wachstumsfaktoren (FGF) umfasst eine Gruppe strukturell verwandter Signalmoleküle, die entscheidende Rollen bei embryonaler Entwicklung, Zellproliferation, Differenzierung, Migration, Angiogenese, Gewebereparatur und metabolischer Regulation spielen. Beim Menschen umfasst die FGF-Familie 18 kanonische Mitglieder (FGF1–FGF10 und FGF16–FGF23), die basierend auf Sequenzhomologie und funktionellen Eigenschaften in sechs Unterfamilien eingeteilt werden. Während die meisten FGFs lokal in einem parakrinen Modus wirken, funktioniert die FGF-19-Unterfamilie (FGF19, FGF21 und FGF23) endokrin und reguliert den systemischen Stoffwechsel von Gallensäuren, Glukose und Phosphat. FGFs entfalten ihre Wirkung durch Bindung an FGF-Rezeptoren (FGFR), wodurch sie nachgelagerte Signalwege wie MAPK und PI3K/AKT aktivieren und damit verschiedene zelluläre Prozesse beeinflussen.
